DRUG INTERACTIONS OF TAZZLE TABLET

Concurrent use increased the risk of hypotension with ± blockers. So, increased risk of priapism with other medications for erectile dysfunction, e.g., alprostadil; increased heart price with theophylline; composed tadalafil serum concentration with CYP 3A4 inducers, e.g., rifampicin, efavirenz, carbamazepine, nevirapine, barbiturates, phenobarbital, phenytoin, rifabutin; increased tadalafil serum concentration with CYP3A4 inhibitors, e.g., azole antifungals, protease inhibitors, cimetidine, macrolides. Potentially Fatal: Enhanced hypotensive effect with nitrates and nicorandil.

CONTRAINDICATIONS OF TAZZLE TABLET

Concurrent use of natural nitrates and nitric oxide donators. So, individuals with cardiac syndrome for whom the sensual activity is improper. So, recent MI (within 90 days) or stroke (within last six mth), hypotension (<90/50 mm Hg), volatile angina, heart failure, independent arrhythmias, or hypertension.

MECHANISM OF ACTION OF TAZZLE TABLET

Tadalafil is a phosphodiesterase type-5 inhibitor. So, onset: 30 min Duration: Up to 36 hr. Union: Well absorbed. So, peak plasma groups entered within 2 hr. And, the rate and extent of conversion not affected by food. Distribution: Widely distributed into tissues. 94% bound to plasma proteins. Metabolism: Metabolised in the organ mainly by the cytochrome P450 isoenzyme CYP3A4 to inactive metabolites. The mean half-life is 17.5 hr. Excretion: Excreted mainly as metabolites in the feces (61% of the dose) and, to a lesser amount, the urine (36% of the amount).

SPECIAL PRECAUTIONS FOR TAZZLE 10MG TABLET

Hepatic or renal impairment; CV diseases; anatomical penile deformation; predisposition to priapism; child <18 yr. Discontinue and seek medical advice if there is swift vision loss or modified vision in one or both eyes or tinnitus, dizziness, sudden loss, or decreased performance while taking Tadalafil. And, seek immediate medical advice if erecting lasts> 4 hr.
